Effect of Low-Intensity Pulsed-Ultrasound Therapy to Increase Osteoblast and Callus Thickness in Fracture Healing

Abstract

Objective: To compare the therapeutic effect of conventional low-intensity pulsed-ultrasound between the control group, treatment group of USD 5x/week and USD 3x/week to the number of osteoblasts and callus thickness in the tibial fracture healing process. Methods: 34 male Rattus norvegicus were divided into 3 groups (control, USD 5x/week, and USD 3x/week). The right tibia was fractured in the middle third, 48 hours later stimulation of ultrasound waves (1 MHz, pulsed, 20% duty cycle, intensity 0.2 W/cm2, duration 10 minutes, stationary) was started in the group USD 3x/week and 5x/week for 3 weeks. After 3 weeks of treatment, all animals were sacrified and callus tissue from the right tibia was used in histological analysis. Results: There were significant differences (p<0.05) in the average number of osteoblasts between the control and USD 5x/week group, between the control and USD 3x/week group, and between USD 5x/week and USD 3x/week group. There were no significant differences (p>0.05) in the callus thickness between all groups. Conclusion: Low-intensity pulsed-ultrasound therapy either frequency 5x/week or 3x/week increased the number of osteoblasts but did not cause a significant increase in the callus thickness at the end of the third week post-intervention on the tibial fracture healing process of Rattus norvegicus.
