Exercise Formula to Induce Beiging Process: A Study Based on Acute Response of Irisin

Abstract

Abstract: Background: Recently, one of treatment approach in obesity is focused on beiging process in white adipose tissue. Exercise has been proposed as one of treatment for obesity, because it could mediate beiging process by means of irisin. Irisin is myokine which is secreted as acute response to exercise. However, the exercise formula which could induce optimal beiging process and lead to an increased level of irisin in high fat diet fed subjects has not been elucidated yet. Objective: This study was aimed to determine the optimal acute response of irisin level and the different response of tested exercise formula between rats fed with standard diet and high fat diet rats. Methods: This study design was in vivo experimental study using 24 Male Sprague Dawley Rats ( 8-10 weeks in age). Rats were grouped into 4 groups (n=6): (1) Standard Diet-HIIE group (SH) (2) Standard diet-CME group (SC) (3) High fat diet (HFD)-HIIE group (HH) (4) High fat diet (HFD)-CME group (HC). After 10 week-diet induction, SH and HH groups were submitted to high intensity intermittent exercise (HIIE), while SC and HC groups were administered to continuous moderate exercise (CME). Blood was collected immediately after exercise. Irisin levels were measured by using ELISA method. Results: This study showed that there were significant differences in acute responses of irisin levels among groups (p=0.035, p<0.05). In addition, it showed that the acute responses to different formula of exercise between standard diet fed rats and high fat diet fed rats were different. Conclusion: Circulating irisin level was affected by exercise formula and individual metabolic status. In HFD-fed subjects, HIIE (high intensity intermittent exercise) formula was better in improving circulating irisin level than CME, while in normal metabolic state, CME (continuous moderate exercise) induced the greater response.
