Abstract

Abstract: Diabetes mellitus is a lifelong, metabolic disease that is characterized by an inability to maintain normal glucose homeostasis. To study the underlying mechanism of diabetes as well as to identify and discover novel treatments, animal models are increasingly being used. However, in Indonesia there are only a small number of study aimed to find the best regimen to create diabetic animal models. Our goal is to find the best regiment to create diabetic animal model. Fat diet combined with different dosages of Streptozotocin (STZ) was used. Twelve adult Sprague-Dawley rats were used in this preliminary study. Fat diet (FT) was given for 10 weeks. The rats were divided into 4 groups, each group was injected intraperitoneally with a dosage of 30, 35, 40 and 50 mg kgBW-1 of STZ. Three to seven days after the injection, blood glucose was measured and compared. The result showed that administration of 30 mg kgBW-1 STZ was sufficient to create rat diabetic animal model which was established 3-7 days after the injection.
