Effect of Exhaustive Exercise on Blood Lymphocyte Count and Diameter of Splenic White Pulp in Rats

Abstract
Abstract: Background. Exhaustive exercise can have negative impacts on the immune system through the oxidative stress pathway. Objectives. The aim of this study was to determine the effects of prolonged exhaustive exercise on the level of malondialdehyde (MDA), the blood lymphocyte count, and the diameter of splenic white pulp. Methods. This study was a true laboratory experiment with a post-test-only control group design. Twenty-eight male rats (Rattus norvegicus) of the Wistar strain were divided into four groups: a control group of 15 days (K1), a treatment group of 15 days (P1), a control group of 30 days (K2), and a treatment group of 30 days (P2). Treatment was given as exhaustive swimming with a load of 6% of bodyweight on the tail. Twenty-four hours after the last treatment, blood was collected to determine the level of MDA and blood lymphocyte count. The animals were sacrificed and their spleens were removed and processed using histological preparation with hematoxylin and eosin (H&E) staining. The diameter of white pulp was measured using a light microscope with 100x magnification using Motic Image Plus 2.0 ML software. Data were analyzed using one-way Analysis of Variance (ANOVA) with a significance level of 0.05. Results. Blood MDA level increased significantly in the P1 and P2 groups. The diameter of white pulp decreased significantly in the P1 group, and lymphocytosis occurred in the P2 group. Conclusion. Prolonged exhaustive exercise leads to oxidative stress and modulates the immune system in animal studies.
